ASSA13-03-8 Effects of Bone Marrow Mesenchymal Stem Cells Transplantation on MMP-2, MMP-9 and TIMP-1 in Heart Failure Rats Myocardium

Abstract

Objective To study the effects of bone marrow mesenchymal stem cells (BMSCs) transplantation on matrix metalloproteinases-2(MMP-2), matrix metalloproteinases-9 (MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in heart failure rats myocardium.

Methods Heart failure model was established by consecutively subcutaneous injection of isoproterenol in wistar rats. These rats were divided 3 groups randomly (8 rats each group). 3 groups were implanted bone marrow mesenchymal stem cells, bone marrow mesenchymal stem cells conditioned medium and saline respectively. At 6 weeks after transplantation, the RT-PCR was used to observed the gene expression of MMP-2, MMP-9, TIMP-1 and the protein expression of MMP-2, MMP-9 and TIMP-1 were measured by Western-blot analysis in heart failure rats myocardium, the protein expression of NF-κB p65, p50 in NE and CE were also measured by Western-blot analysis, and the protein expression of IκB, p-IκB in total myocardium protein were also measured by Western-blot analysis.

Results Compared with the saline group, the gene and protein expression of MMP-2, MMP-9 in BMSCs group and BMSCs-CM group were less singnificantly, P < 0.05; but the gene and protein expression of TIMP-1 in BMSCs group and BMSCs-CM group was more obviously than the saline group, P < 0.05; the gene and protein expression of MMP-2, MMP-9 in BMSCs group were more singnificant than BMSCs-CM group, P < 0.05; but the gene and protein expression of TIMP-1 in BMSCs group was more obviously than BMSCs-CM group, P < 0.05; compared with BMSCs group and BMSCs-CM group, the expression of NF-κB p65 and p50 in NE in saline group were more singnificant, P < 0.05; however, the expression of NF-κB p65 and p50 in CE in saline group were less than BMSCs group and BMSCs-CM group, P < 0.05; compared with BMSCs–CM group, the protein expression of NF-κB p65 and p50 in NE in BMSCs group were more, P < 0.05; but the protein expression of NF-κB p65, p50 in CE in BMSCs group were less than BMSCs group, P < 0.05; the value of p-IκB/IκB was different in 3 groups and in the term of the value of p-IκB/IκB, the saline group was the highest and the BMSCs group was the lowest in 3 groups, P < 0.05.

Conclusions The transplantation of BMSCs and BMSCs-CM both can increase the level of MMP-2 and MMP-9 and decrease the level of TIMP-1 at same time in heart failure rats myocardium. its maybe mechanism is that BMSCs and BMSCs-CM both can change the number of NF-κB in NE by inhibiting the activity of IκB in heart failure rats myocardium, and the decrease of NF-κB in NE was reduced to influence gene transplantation.
