A rapid and potent natriuretic response to intravenous injection of atrial myocardial extract in rats

Abstract

Supernatants of atrial or ventricular myocardial homogenates were injected intravenously into anaesthetized non-diuretic rats. Extract derived from atrial muscle caused a rapid, more than 30-fold increase of sodium and chloride excretions, while urine volume rose 10-fold, and potassium excretion doubled. No such changes were observed after injection of ventricular tissue extract obtained from the same rat hearts, or after injection of the homogenization medium. There was no significant difference in glomerular filtration rates between the two groups. We conclude that the atrial extract contained an extremely powerful inhibitor of renal tubular NaCl reabsorption.

It is becoming widely understood that gut microbiota and human health are related. It is now well-accepted that healthy gut flora plays a significant role in the host's overall health. The gut flora is a diverse and dynamic collection of microorganisms in the human gastrointestinal (GI) tract, significantly impacting the host during homeostasis and disease. This microbial community's diversity is host-specific and changes throughout an individual's lifespan. The gut flora controls several metabolic pathways in the host, leading to interacting host-microbiota metabolic, signalling and immune-inflammatory axes that physiologically link the gut with the brain, heart, lung and skin. Numerous inflammatory illnesses and infections have been connected to altered gut bacterial composition or dysbiosis. Optimising therapeutic and probiotic approaches to control the gut microbiota to treat disease and promote health requires a deeper understanding of these axes. This review confers our current understanding of the connections between gut flora with the brain, heart, lungs, and skin and also portrays the diseases correlated with these axes.

Heart failure (HF) is a complex clinical syndrome associated with high rates of morbidity and mortality. Over the years, it has been crucial to find accurate biomarkers capable of doing a precise monitor of HF and provide an early diagnosis. Of these, it has been established an important role of natriuretic peptides in HF assessment. Moreover, the development of biosensors has been garnering interest as new diagnostic medical tools. In this review we first provide a general overview of HF, its pathogenesis, and diagnostic features. We then discuss the role of natriuretic peptides in heart failure by characterizing them and point out their potential as biomarkers. Finally, we adress the evolution of biosensors development and the available natriuretic peptides biosensors for disease monitoring.
Developmental roles of natriuretic peptides and their receptors

Natriuretic peptides and their receptors are implicated in the physiological control of blood pressure, bone growth, and cardiovascular and renal homeostasis. They mediate their action through the modulation of intracellular levels of cGMP and cAMP, two second-messengers that have broad biological roles. In this review, we briefly describe the major players of this signaling pathway and their physiological roles in the adult, and discuss several reports describing their activity in the control of various aspects of embryonic development in several species. While the core components of this signaling pathway are well conserved, their functions have diverged in the embryo and the adult to control a diverse array of biological processes.
Endocrine and scavenging cell functions of a juvenile fish heart atrium: Fine structure aspects and translating conjectures

The atrium of the juvenile teleost, smallmouth bass, Micropterus dolomieu Lacépède showed a pigmented endocardium. Our goal was to use fine structure to survey this fish heart tissues and verify the content responsible for this shrouding and discuss mammalian, translational conjectures about this cardiac structure.
Using electron microscopy, heart pericardium, myocardium, and endocardium were analyzed, including atrial peptide immunolabeling.
The endocardium endothelial cells of the atrium revealed pinocytosis and endocytosis activities that resulted in accumulated electron-contrasted secondary lysosomes and lipofuscin bodies not found in the ventricle endocardium. This endothelium contacted subjacent atrial and ventricular myocardial cells producing immunolabeled atrial peptide-containing vesicles. Other migrating cells, including melano-macrophages, were noticed in the atrial sub endocardium.
The endocardium functioned as a potent blood-heart barrier where transcytosis and dispatching of numerous materials, including those atrial peptides, between myocardium and the circulation occur. Peculiarly, this juvenile endothelium revealed abundant lysosomal processing of scavenged circulation materials like in a reticuloendothelial tissue. These juvenile cell's accumulated ‘aging’ lipofuscin bodies as organized deposits into a network with other organelles, especially mitochondria without evident disposal. Additionally, other cells, including melano-macrophages roamed both sub endocardium and ventricle; those also can influence the viewed pigmentation. Some of these data comforted the endocardial mesodermal lineage, still sketchy for all vertebrates while fish heart development is still used. These observations could raise questions: Does turnover of lipofuscin and associated captures make endocardial cells to model other mammalian pathologies, including those associated with heart failure?
