2020, Clinical ImmunologyCitation Excerpt :In view of our present data, the exposure of M2R to anti-M2R Ab should lead to the attenuation of negative inotropism as a result of the inhibition of agonist-induced, Gi2 protein-mediated signaling. In fact, previous studies have shown that pretreatment of isolated rat atria preparations with DCD IgG, its corresponding F(ab’)2 fragment or monospecific IgG antibodies against the M2R's II-ECL result in an a noncompetitive inhibition of agonist-induced negative inotropic effects in isolated rat atria [11,12,32]. As regards the ability of anti-M2R Ab to promote inhibition of agonist-mediated M2R activation, two potential mechanisms could explain this effect.

But at the same time, the presence of autoantibodies against muscarinic M2 receptors has been detected that may be the cause of the failure in adrenergic activity [47,48]. Interestingly, Cremashi et al. [49] have suggested that the existence of cross-reactivity between a parasite antigen and the major antigenic epitopes present on both, β1-adrenergic and M2-cholinergic receptors, may explain the modulatory effect on the myocardial physiology.

The second extracellular loop was selected by some authors [111], on the grounds that it is an epitope of anti-receptor antibodies for many G-protein coupled receptors in other autoimmune conditions such as Chagas cardiomyopathy [116], idiopathic dilated cardiomyopathy [117] and neonatal lupus [118]. Three overlapping peptides spanning the entire length of the second extracellular loop of M3R were synthesized and rabbits were immunized with a mixture of them, emulsified in complete Freund's adjuvant.
