Elafin is a potent inhibitor of proteinase 3

Abstract

Elafin, a human skin derived inhibitor of human leukocyte elastase, was tested for inhibitory activity against proteinase 3, an elastin degrading proteinase of neutrophils. The inhibitory activity of elafin was compared with antileukoprotease and eglin C. Elafin proved to be a potent inhibitor of elastin-FITC degradation showing an IC 50 of 9.5 × 10−9 M. Potency was found to be more than 100-fold higher as compared with antileukoprotease and eglin C.

    SLPI, a well-studied WAPD protein with antiprotease and antibacterial activities, prevents inflammation via inhibiting proteases released from neutrophils and protects the host against different kinds of pathogens [7–9].     Despite elafin shares 40% sequence identity with SLPI, the protease inhibitory domain is identical except for the P1 residue (Ala for elafin vs Leu for SLPI). This single mutation may explain why elafin has a more specific spectrum of inhibition than SLPI [69], since elafin inhibits proteinase 3 (Pr3) but not trypsin, chymotrypsin or Cat G [70,71]. Elafin displays potent antibacterial and antifungal activities against P. aeruginosa and S. aureus as well as C. albicans and A. fumigatus.
